Synopsis
 

The current proposal is built on a hospital based case-control study conducted at Tata Memorial Hospital, Mumbai in the Year 2009-2017.The study completed the first genome-wide analysis (GWAS) of gallbladder cancer; conducted extensive analyses related to lifestyle factors; identified immunological prognostic markers, and analyzed the role of H.Pylori. Important lifestyle factors identified were: high consumption of mustard oil, high consumption of freshwater fish and central obesity. Given the potential of these lifestyle factors to reduce the burden of GBC, an urgent replication of findings is planned by establishing a multicentre case-control study in the GBC high incident region of India. our GWAS and replication analyses provide substantial evidence for the association between common genetic variation in the ABCB1 and ABCB4 gene regions and risk of gallbladder cancer. It is possible that identified genetic factors interact with lifestyle factors. Heritability analysis indicates the likely existence of many more common variants associated with gallbladder cancer that could be discovered with a larger sample set than the existing case-control study. The overall aim of the current proposal is to replicate findings of TMH study and to establish a platform to conduct laboratory analysis for GWAS, detection of heavy metals and identification of the role of S. Typhi in gallbladder cancer using the standardized protocol on a large number of samples. The study will enroll 3000 cases and similar number controls. The study will collect lifestyle questionnaires, anthropometric measurements, blood samples, tissue samples(whenever possible), clinical data and other information from study participants from 5 study centers.
